Tri (2-carboxyethyl) phosphine hydrochloride (TCEP · HCl) is a thiol free and odorless compound, which is widely used in biochemistry and molecular biology as a disulfide bond reducing agent for peptides or proteins. With other similar reducing agents such as dithiothreitol (DTT), β- Mercaptoethanol( β- Compared with ME), TCEP · HCl has many advantages:

1) No odor - it can be used on the test bench without fume hood;

2) High stability - the stability of the structure of the compound itself makes it unnecessary to take any preventive measures to avoid oxidation during operation, use and preservation. It will not react with other functional groups in the protein;

3) Wide pH working range - it is almost soluble in water soluble buffer of any pH value, and the effective working range of pH is 1.5-9.0. The pH value after directly dissolving in water is about 2.5;

4) Strong reducibility - For most experiments, 5-50 mM TCEP · HCl can provide enough molar force to fully reduce the disulfide bond by acting at room temperature for a few minutes;

5) Wide compatibility - it does not contain mercaptan group, so it is not necessary to remove this reductant in advance for downstream mercapto modification experiments such as maleimide coupling.

This product is suitable for the preparation steps of gel electrophoresis and solid phase metal ion affinity chromatography (IMAC). It is used as a reducing agent to destroy disulfide bonds within or between proteins. It is also particularly suitable for the reaction of maleimide coupling cysteine residues. It can prevent cysteine residues from forming disulfide bonds, and is not similar to DTT or β- ME itself is easy to react with maleimide. RNA isolation experiments are also commonly used in the tissue homogenate step.
